Anti-Apoptotic Effects Of Valproic Acid Treatment On Dopaminergic Neuronal Loss In A 6-Hydroxydopamine Model Of Parkinson’s Disease In Rats

Aim: Parkinson’s disease (PD) is characterized by the progressive loss of dopaminergic neurons resulting in deterioration motor activity patients. Currently, available therapies including Levodopa (L-DOPA) are more geared toward treatment symptoms. Therefore, developing effective neuroprotective needed. Valproic acid (VPA) has shown potent effects on dopamine (DA) various brain regions. The aim this study to investigate whether VPA attenuates neuronal when co-treated with L-DOPA a 6-hydroxydopamine (6-OHDA) induced PD model rats.Methods: Male Wistar Albino rats received intranigral injection 6-OHDA unilaterally. Twelve days later either saline, L-DOPA, VPA, or L-DOPA+ for 9 days. To determine had apomorphine-induced rotation test was used. Immunohistochemical analyses were performed Substantia Nigra pars compacta (SNpc) measuring tyrosine hydroxylase (TH) positive and apoptotic neurons.Results: showed clinically impairment function histologically significant damage neurons. administration combined L- DOPA protected SNpc increasing TH decreasing given as monotherapy, other hand, ineffective these parameters.Conclusion: Our experiments demonstrated that effect used rat model.
